Coumarin compounds represent a diverse class of natural and synthetic molecules with significant therapeutic potential. This comprehensive review examines the therapeutic landscape of coumarin compounds across multiple disease areas, integrating detailed mechanism-of-action analysis with translational context. Natural coumarins and their synthetic derivatives are discussed, with focus on key pharmacological activities including antimicrobial, anti-inflammatory, anticancer, and antioxidant properties. Structure-activity relationships are analyzed to identify therapeutic optimization strategies. The review provides evidence-driven insights for researchers working on coumarin-based drug discovery and development.
